Biography

Dr. De received Bachelor of Science degree with honors in chemistry and Master of Science in biochemistry from Calcutta University, India. He worked for few years at a National Cancer Institute in India before coming to US for post- graduate education in 1975. After completion of Ph.D in biochemistry from University of Maryland in 1980, he underwent two postdoctoral trainings in basic cancer research under the supervisions of Dr. Leon Heppel, National Academy of Sciences, at Cornell University, Ithaca, NY and Dr. Stanley Cohen, Nobel laureate, Medicine 1986, at Vanderbilt University, Nashville, TN. Gradually, he became highly interested in Clinical Chemistry. In 1993, he took a fellowship in clinical chemistry and toxicology under the supervisions of both Dr. Amadeo Pesce and Dr. Bradley Copeland at University of Cincinnati Medical Center, Cincinnati, OH. Following completion he joined University of Mississippi Medical Center in 1995 as the Associate Director of clinical chemistry and toxicology. In 2005, Dr. De was appointed the Director of clinical chemistry and toxicology in the department of Pathology, University of Arizona Health Sciences Center. He is a board certified clinical chemist from the American Board of Clinical Chemistry, a board certified toxicologist from the National Registry of Clinical Chemistry and a fellow of the National Academy of Clinical Biochemistry. Since 2010, Dr. De has been serving as a member of the board of directors in the American Board of Clinical Chemistry, Toxicology and Molecular Biology. He was nominated to be the chair of nominating committe of this board in 2015. He has published more than 70 peer reviewed articles and meeting abstracts in basic and clinical sciences. He is a reviewer of both national and international journals and received recognitions as a Clinical Chemist from the American Association of Clinical Chemistry. In 2013 he was honored for the “Paul Finley Chemist Award” from University of Arizona Health Sciences Center. He is the co-principal investigator of a Center of Disease Control (CDC) sponsored project on HIV screening and diagnosis. In 2019 he served as a regional president (Rocky Mountain Section) of the American Board of Clinical Chemistry.

Tacrolimus, an immunosuppressive drug discovered in 1984, is used to decrease the risk of organ rejection. The 2007 European Consensus Conference on Tacrolimus Optimization recommended the use of methods with a limit of quantitation at < or =1ng/ml for monitoring low dose tacrolimus therapy.

Measurements of serum iron and total iron-binding capacity (TIBC) can be used to aid the diagnosis of iron deficiency and iron overload states. A variety of different methods for these measurements are commercially available..Linearity, imprecision and hemoglobin interference of homogeneous iron and TIBC methods on a Dimension RxL analyzer were assessed. C-reactive protein (CRP) is a nonspecific marker of inflammation that can be used as a marker for atherosclerotic risk. This application requires increased precision at low CRP concentrations compared to traditional assays.

Glycohemoglobin (gHb) is a marker of long-term glycemic control that has been shown to correlate with complications of diabetes mellitus (1). The National Glycohemoglobin Standardization Program (NGSP) was established to standardize gHb results so that clinical laboratory results are comparable to those reported by the Diabetes Control and Complications Trial (2)(3). Previous studies have shown that some gHb methods yield inaccurate results with samples heterozygous for hemoglobin (Hb) C or Hb S (4)(5)(6). At least 10% of black Americans have either Hb C or S trait, and there were 19 million black Americans over age 19 in 1990 (7)(8)(9)(10). The prevalence of diabetes is estimated to be 5.1% of the adult population, with the rate for non-Hispanic blacks being 1.6-fold higher than that of non-Hispanic whites (11). It is therefore probable that at least 150 000 Americans with diabetes have Hb C or S trait. Here, we investigate the measurement of gHb in specimens containing Hb C or S trait using eight gHb methods currently in clinical use. Whole blood samples from individuals homozygous for Hb A (n = 43) and heterozygous for Hb C or S (n = 43 and 61, respectively) were collected in EDTA tubes. Hb variants were identified by their characteristic Variant A1c HPLC chromatograms (Bio-Rad Laboratories). Aliquots of these samples containing between 4% and 14% Hb A1c (NGSP Hb A1c evaluation range) were stored refrigerated (2–8 °C) and analyzed within 10 days of collection. This study was approved by the Institutional Review Board of the University of Utah. The CLC 330 gHb analyzer (Primus Corporation) was operated at the University of Missouri.
